"RXRβ is one of three types of retinoid X receptors, which play important roles in how cells grow, differentiate, and die and might be targets for the treatment of conditions such as insulin resistance, autoimmunity, and neurodegeneration. A recent study into the physical behavior of RXRβ could bring researchers closer to that possibility. A combination of lab experiments and computer modeling revealed the unique properties of the receptor’s AB region. This region, common to this family of receptors, enables the activation of target genes. But in RXRβ, researchers found, the AB region also supports liquid-liquid phase separation a biochemical phenomenon that is fundamental to the compartmentalization of the cell. As a driver of RXRβ’s physical behavior, this capacity for phase separation could also influence the receptor’s transcriptional behavior. Understanding how could give researchers a better idea of RXRβ’s role in disease and how it might be modulated to promote human health..."

"Phospholipid scramblase 1 (PLSCR1) is the most studied member of its protein family and has a complex array of suggested functions. While PLSCR1 was first described as a plasma membrane protein, it has subsequently been found in other cellular compartments and implicated in numerous cellular pathways including apoptosis, intracellular trafficking of membrane proteins, and pro-inflammatory events. PLSCR1 interacts with various effectors, mediators, and regulators that then contribute to distinct cellular processes. While most PLSCR1 interactors are thought to be cell-type specific, the mechanisms of PLSCR1's regulatory functions are often shared. But not all PLSCR1 interactors are endogenous in origin; PLSCR1 also interacts with exogenous viral proteins. These interactions regulate viral uptake and spread in both pro- and anti-viral ways..."

"The hormone and receptor pair IGF-1 and IGF-1R and the Hippo-YAP signaling pathway are critical to breast cancer stem cells, but if or how these pathways interact in these cells remained unclear. So, researchers recently examined YAP and IGF-1R in triple negative breast cancer cell lines and xenograft models. YAP knockdown reduced viability and stemness in cell culture and tumorigenicity in vivo, suggesting that YAP contributes to stemness in breast cancer cells. Further tests determined that IGF-1R regulated YAP expression, and in turn, YAP regulated IGF-1 expression but not IGF-1R. Specifically, depleting IGF-1R decreased YAP expression, while addition of IGF-1 upregulated YAP and increased its nuclear localization. YAP overexpression increased IGF-1 expression without impacting IGF-1R expression. In clinical data from basal-like breast cancer patients, higher levels of YAP and IGF-1 are correlated with shorter overall survival..."
